    A Data Structure for Spatio-Temporal Databases

    The advantages and applications of spatial mechanisms are well documented; however, there are very few being designed. The principal hinderance to the design of spatial mechanisms is the great difficulty involved in specifying spatial problems and in interpreting spatial solutions. Similarly, the development of spatial codes to implement these techniques is held back by the lack of means to easily visualize and verify solutions, particularly in the realm of relational databases. If spatial mechanisms are to be successful, the designer must be able to synthesize, analyse and evaluate, as well as load and extract information, using a single code representing a spatial structure. This entails the implementation of spatial relationships involving spatial data structures. It is with this in mind that the Canadian Hydrographic Service database group embarked on the development of a new type of spatial database structure based on the quadtree concept

    View the Executive SummaryIncreased reliance on the Internet and other networked systems raise the risks of cyber attacks that could harm our nation’s cyber infrastructure. The cyber infrastructure encompasses a number of sectors including: the nation’s mass transit and other transportation systems; banking and financial systems; factories; energy systems and the electric power grid; and telecommunications, which increasingly rely on a complex array of computer networks, including the public Internet. However, many of these systems and networks were not built and designed with security in mind. Therefore, our cyber infrastructure contains many holes, risks, and vulnerabilities that may enable an attacker to cause damage or disrupt cyber infrastructure operations. Threats to cyber infrastructure safety and security come from hackers, terrorists, criminal groups, and sophisticated organized crime groups; even nation-states and foreign intelligence services conduct cyber warfare. Cyber attackers can introduce new viruses, worms, and bots capable of defeating many of our efforts. Costs to the economy from these threats are huge and increasing. Government, business, and academia must therefore work together to understand the threat and develop various modes of fighting cyber attacks, and to establish and enhance a framework to assess the vulnerability of our cyber infrastructure and provide strategic policy directions for the protection of such an infrastructure. This book addresses such questions as: How serious is the cyber threat? What technical and policy-based approaches are best suited to securing telecommunications networks and information systems infrastructure security? What role will government and the private sector play in homeland defense against cyber attacks on critical civilian infrastructure, financial, and logistical systems?     Gain properties of dye-doped polymer thin films

    Hybrid pumping appears as a promising compromise in order to reach the much coveted goal of an electrically pumped organic laser. In such configuration the organic material is optically pumped by an electrically pumped inorganic device on chip. This engineering solution requires therefore an optimization of the organic gain medium under optical pumping. Here, we report a detailed study of the gain features of dye-doped polymer thin films. In particular we introduce the gain efficiency KK, in order to facilitate comparison between different materials and experimental conditions. The gain efficiency was measured with various setups (pump-probe amplification, variable stripe length method, laser thresholds) in order to study several factors which modify the actual gain of a layer, namely the confinement factor, the pump polarization, the molecular anisotropy, and the re-absorption. For instance, for a 600 nm thick 5 wt\% DCM doped PMMA layer, the different experimental approaches give a consistent value K≃K\simeq 80 cm.MW−1^{-1}. On the contrary, the usual model predicting the gain from the characteristics of the material leads to an overestimation by two orders of magnitude, which raises a serious problem in the design of actual devices. In this context, we demonstrate the feasibility to infer the gain efficiency from the laser threshold of well-calibrated devices.     Semisynthetic Nanoreactor for Reversible Single-Molecule Covalent Chemistry

    Protein engineering has been used to remodel pores for applications in biotechnology. For example, the heptameric alpha-hemolysin pore (alpha HL) has been engineered to form a nanoreactor to study covalent chemistry at the single -molecule level. Previous work has been confined largely to the chemistry of cysteine side chains or, in one instance, to an irreversible reaction of an unnatural amino acid side chain bearing a terminal alkyne. Here, we present four different alpha HL pores obtained by coupling either two or three fragments by native chemical ligation (NCL). The synthetic alpha HL monomers were folded and incorporated into heptameric pores. The functionality of the pores was validated by hemolysis assays and by single-channel current recording. By using NCL to introduce a ketone amino acid, the nanoreactor approach was extended to an investigation of reversible covalent chemistry on an unnatural side chain at the single -molecule level

    Caffeic Acid Phenethyl Ester and Its Amide Analogue Are Potent Inhibitors of Leukotriene Biosynthesis in Human Polymorphonuclear Leukocytes

    BACKGROUND: 5-lipoxygenase (5-LO) catalyses the transformation of arachidonic acid (AA) into leukotrienes (LTs), which are important lipid mediators of inflammation. LTs have been directly implicated in inflammatory diseases like asthma, atherosclerosis and rheumatoid arthritis; therefore inhibition of LT biosynthesis is a strategy for the treatment of these chronic diseases. METHODOLOGY/PRINCIPAL FINDINGS: Analogues of caffeic acid, including the naturally-occurring caffeic acid phenethyl ester (CAPE), were synthesized and evaluated for their capacity to inhibit 5-LO and LTs biosynthesis in human polymorphonuclear leukocytes (PMNL) and whole blood. Anti-free radical and anti-oxidant activities of the compounds were also measured. Caffeic acid did not inhibit 5-LO activity or LT biosynthesis at concentrations up to 10 ”M. CAPE inhibited 5-LO activity (IC(50) 0.13 ”M, 95% CI 0.08-0.23 ”M) more effectively than the clinically-approved 5-LO inhibitor zileuton (IC(50) 3.5 ”M, 95% CI 2.3-5.4 ”M). CAPE was also more effective than zileuton for the inhibition of LT biosynthesis in PMNL but the compounds were equipotent in whole blood. The activity of the amide analogue of CAPE was similar to that of zileuton. Inhibition of LT biosynthesis by CAPE was the result of the inhibition of 5-LO and of AA release. Caffeic acid, CAPE and its amide analog were free radical scavengers and antioxidants with IC(50) values in the low ”M range; however, the phenethyl moiety of CAPE was required for effective inhibition of 5-LO and LT biosynthesis. CONCLUSIONS: CAPE is a potent LT biosynthesis inhibitor that blocks 5-LO activity and AA release. The CAPE structure can be used as a framework for the rational design of stable and potent inhibitors of LT biosynthesis

    Factors influencing mentors’ satisfaction: A study from medical schools in Norway and Canada

    Phenomenon: The mentoring of undergraduate medical students has been shown to benefit the mentors; however, detailed information on the factors that influence the satisfaction and motivation of mentors remains unclear. Such knowledge can be useful in sustaining group mentorship programs. The aim of this study was to investigate the experiences and perspectives of mentors to ascertain the factors that contribute to satisfaction and motivation. Approach: As part of a larger research project, a survey was sent out to mentors at UiT the Arctic University of Norway, the University of Bergen and McGill University (N=461). Descriptive statistics, linear regression and factor analyses were used to examine the data in order to map factors associated with mentor satisfaction. Findings: The overall response rate was 59% (n=272/461). Mentors reported a high mean satisfaction score of 4.55 (±0.04, median 5.00) on a five-point Likert scale. Six out of nine statements describing how mentors approach group mentoring were strongly correlated with each other. Through factor analysis of the items, we found a dominating factor labeled “Student-centered mentoring approach” which was strongly associated with the level of satisfaction as a mentor. Additionally, highly satisfied mentors took a greater interest in patient-centered medicine and their students’ personal development. Their groups spent more time discussing students’ clinical experiences, societal poverty and health, and patients’ suffering and sickness. Insights: Our findings suggest that high mentor satisfaction, which is important for the pedagogical quality and sustainability of mentor programs, is related to the mentors’ student-centeredness and their interest in topics concerning professionalism. By preparing mentors for their roles and supporting them in developing strategies for establishing good mentoring relationships, the outcomes of group mentoring may be improved both for mentors and students. Interest in students’ personal development and the mentors’ own professional development seem to be indicators of mentors’ satisfaction and should be encouraged in mentorship programs

    A variant of sparse partial least squares for variable selection and data exploration

    When data are sparse and/or predictors multicollinear, current implementation of sparse partial least squares (SPLS) does not give estimates for non-selected predictors nor provide a measure of inference. In response, an approach termed "all-possible" SPLS is proposed, which fits a SPLS model for all tuning parameter values across a set grid. Noted is the percentage of time a given predictor is chosen, as well as the average non-zero parameter estimate. Using a "large" number of multicollinear predictors, simulation confirmed variables not associated with the outcome were least likely to be chosen as sparsity increased across the grid of tuning parameters, while the opposite was true for those strongly associated. Lastly, variables with a weak association were chosen more often than those with no association, but less often than those with a strong relationship to the outcome. Similarly, predictors most strongly related to the outcome had the largest average parameter estimate magnitude, followed by those with a weak relationship, followed by those with no relationship. Across two independent studies regarding the relationship between volumetric MRI measures and a cognitive test score, this method confirmed a priori hypotheses about which brain regions would be selected most often and have the largest average parameter estimates. In conclusion, the percentage of time a predictor is chosen is a useful measure for ordering the strength of the relationship between the independent and dependent variables, serving as a form of inference. The average parameter estimates give further insight regarding the direction and strength of association. As a result, all-possible SPLS gives more information than the dichotomous output of traditional SPLS, making it useful when undertaking data exploration and hypothesis generation for a large number of potential predictors. © 2014 Olson Hunt, Weissfeld, Boudreau, Aizenstein, Newman, Simonsick, Van Domelen, Thomas, Yaffeand Rosano

    Sustainable development in the mining industry: Towards the development of tools for evaluating socioeconomic impact in the Canadian context

    In spite of its economic outputs, the Canadian mining sector has suffered from several setbacks that have compromised its reputation. This has led many of its stakeholders to adopt a shift towards a new paradigm focused on sustainable development. In this review of the literature, we identify the impact that mining activities have had on various social, economic and environmental issues in Canadian society. We also survey the criteria and indicators included in various models that may be used to evaluate the adequacy of the transition of mines towards sustainable development. Finally, the strengths and weaknesses of eight tools that have been applied to the socioeconomic impact of mines are analysed. A clear vision of prospects for promoting sustainable development in Canadian mining emerges from this survey. Our analysis reveals important factors not given adequate consideration in the models used currently to evaluate and suggests opportunities to improve these evaluation tools through identification of more relevant criteria and indicators. The research undertaken should incite stakeholders to develop social and economic impact evaluation tools that are better adapted to the Canadian context
